This Amended and Restated Employment Agreement (this “Agreement”) amends and restates, effective as of the 5th day of February, 2010, that certain Employment Agreement made and entered into as of the 11th day of February, 2008, as amended by that certain Amendment to Employment Agreement entered into as of the 27th day of December, 2008 (collectively, the “Original Agreement”) by and between Vertex Pharmaceuticals Incorporated, a Massachusetts corporation (together with its successors and assigns, the “Company”), and Peter Mueller (the “Executive”).
