Canonical Signaling Pathways Regulated by Gα and Gβγ Subunits. Schematic representation of classical GPCR signaling via Gα and Gßγ subunits. Upon dissociation of the Gßγ subunits from Gα, individual subunits initiate diverse signaling cascades. Gαs is most known for adenylyl cyclase (AC) stimulation leading to an increase in cytosolic cAMP. Conversely, Gαi inhibits AC reducing cytosolic cAMP. Gαq activates phospholipase C (PLC) which results in increased intracellular Ca2+ and activation of PKC, and Gα12/13 activates RhoGEF. Gßγ subunits are most known for their signaling to various ion channels, PLC, AC, PI3K, and MAPK leading to diverse responses within the cell.
