Functional redundancy among CELF protein family members A prominent splicing target for CELF proteins is Cardiac troponin T (cTNT). The alternative splicing pattern of cTNT is developmentally regulated and conserved from birds to mammals (Xxxx et al., 2001). The alternative cTNT exon 5 which is normally included in the embryo of cardiac and skeletal muscle is skipped in adult cardiac muscle mRNA (Xxxx et al., 2001) (Xxxxxxx et al., 2006). Alternative splicing of cTNT exon 5 generates isoforms with different contractive properties important during muscle development (Xxxxxx and Ordahl, 1985). All CELF proteins bind to conserved intronic elements called muscle-specific splicing enhancer elements (MSEs) which contains a CUG repeat to promote exon 5 inclusion (Xxxx et al., 2001). Although this splicing event is dependent on different intronic splicing enhancer (ISE) elements, CELF3 and CELF6 promote exon 5 inclusion in the presence of a muscle-specific ISE element (MSE2) found at the proximal region flanking exon 5 while other CELF proteins require several MSE elements (Xxxx et al., 2001). Furthermore, all regions within CELF proteins are implicated in the splicing regulation of cTNT exon 5. These include sequences within RRM1 and RRM2 of CELF2 as well as regions within the divergent domain and RRM1 of CELF4 (Xxxxx et al., 2004). Altogether, these findings suggest that different ISE elements in addition to domains within CELF proteins are indispensable for splicing activity of the same exon which indicates functional redundancy among CELF family members. An orthologue of CELF3, 4 and 5 in Caenorhabditis elegans, unc-75, is neuron specific and involved in neurotransmission where it regulates neuron-specific alternative splicing (Xxxxx et al., 2003). unc-75 null mutants have a motor phenotype that is rescued by human CELF4 (Xxxxx et al., 2003). These studies illustrate the importance of CELF proteins in maintaining neuronal function. Similarly, the generation of mouse models deficient in specific CELF proteins, has helped to elucidate the role of members of the CELF family during alternative splicing. In mice lacking CELF1, growth is retarded; there is impaired viability and reduced fertility due to disruption of spermatogenesis (Xxxxx et al., 2007). CELF3 knock-out mice did not show any reduction in fertility but similar to CELF1 knock-out mice, they have reduced spermatogenesis (Xxx et al., 2007).   • Technical Redundancy Where an employee's employment is being terminated by the employer by reason of the sale or transfer of the whole or part of the employer's business, nothing in this Agreement shall require the employer to pay compensation for redundancy to the employee if:

  • Redundancy The company is, and will remain during the life of this Agreement, a participating employer in the Redundancy Payment Central Fund Ltd (Incolink) and all employees will be enrolled in the Fund and be entitled to redundancy benefits in accordance with the terms of the Deed. The company shall pay contributions on behalf of each employee into the Incolink Number 1 Fund on a weekly basis, as per the Trust Deed.

  • PERSONNEL REDUCTION Section 1 In the event of layoffs in connection with decreasing the work force, and the recall to work of people so laid off, the following consideration shall govern. Skill and ability as determined by reference to the employee's work record, and length of service shall be the determining factors; however, employees shall be laid off by category of seniority. There shall be three (3) seniority categories: probationary, 1 year to 5 years seniority, and over 5 years seniority. In case of layoff, all employees in the lowest seniority category shall be laid off before proceeding to layoff of anyone in a more senior category. Where skill and ability within a category are approximately equal, length of service shall govern. Employees having the same seniority within a category shall draw lots to determine the order of layoff. No new employees shall be hired until all laid off employees have been given the opportunity to be re-hired. Employees who have been laid off will be offered re-employment in the inverse order of layoffs when they are needed again, provided they are physically qualified and possess sufficient training and experience to perform the duties of the available work. The City shall give laid off employees ten (10) days notice of its intention to rehire. The employees shall within ten (10) days period notify the City of their intention to, or not to, return to the employ of the City, and shall report to work no later than fifteen (15) days from receipt of said notice to rehire. If an employee fails to notify the City within the ten (10) calendar day period of his/her intentions to return to work, or fails to report to work within fifteen (15) calendar days from the date of notice, he/she shall be considered permanently severed from the employ of the City. At the time of a layoff the City shall provide all laid off employees with a complete physical examination. At the time of rehire, the City may require a physical examination prior to the employee's return to duty, and it is expressly understood that any employee found physically unfit to return to duty may be refused re-employment and removed from the employment list. The City shall not be obligated to rehire laid off employees who have been laid off for five (5) or more consecutive calendar years, beginning from the date of layoff.

  • Scope of Service Interconnection Service shall be provided to the Interconnection Customer at the Point of Interconnection (a), in the case of interconnection of the Customer Facility of a Generation Interconnection Customer, up to the Maximum Facility Output, and (b), in the case of interconnection of the Customer Facility of a Transmission Interconnection Customer, up to the Nominal Rated Capability. The location of the Point of Interconnection shall be mutually agreed by the Interconnected Entities, provided, however, that if the Interconnected Entities are unable to agree on the Point of Interconnection, the Transmission Provider shall determine the Point of Interconnection, provided that Transmission Provider shall not select a Point of Interconnection that would impose excessive costs on either of the Interconnected Entities and shall take material system reliability considerations into account in such selection. Specifications for the Customer Facility and the location of the Point of Interconnection shall be set forth in an appendix to the Interconnection Service Agreement and shall conform to those stated in the Facilities Study.

  • Switching System Hierarchy and Trunking Requirements For purposes of routing ECI traffic to Verizon, the subtending arrangements between Verizon Tandem Switches and Verizon End Office Switches shall be the same as the Tandem/End Office subtending arrangements Verizon maintains for the routing of its own or other carriers’ traffic (i.e., traffic will be routed to the appropriate Verizon Tandem subtended by the terminating End Office serving the Verizon Customer). For purposes of routing Verizon traffic to ECI, the subtending arrangements between ECI Tandem Switches and ECI End Office Switches shall be the same as the Tandem/End Office subtending arrangements that ECI maintains for the routing of its own or other carriers’ traffic.

  • Scope of Services The specific scope of work for each job shall be determined in advance and in writing between TIPS Member, Member’s design professionals and Vendor. It is permitted for the TIPS Member to provide a general scope description, but the awarded vendor should provide a written scope of work, and if applicable, according to the TIPS Member’s design Professional as part of the proposal. Once the scope of the job is agreed to, the TIPS Member will issue a PO and/or an Agreement or Contract with the Job Order Contract Proposal referenced or as an attachment along with bond and any other special provisions agreed by the TIPS Member. If special terms and conditions other than those covered within this solicitation and awarded Agreements are required, they will be attached to the PO and/or an Agreement or Contract and shall take precedence over those in this base TIPS Vendor Agreement.

  • Employment Categories (a) Employees under this Agreement will be employed in one of the following categories:

  • Overload Compensation ‌ An instructional faculty member may be given overload compensation for additional instructional responsibilities beyond the normal work day or work year. The conditions and compensation of all overload duties shall be established prior to the beginning of additional instructional work. The rate of compensation shall be paid on a prorated salary basis. When calculating the prorated hourly salary under this section, the academic year salary shall be divided by 1344 hours. Faculty members have the option of refusing additional work assignments: when this occurs the rate of compensation will be agreed upon between the employer and the faculty member.

  • JOB All vacant positions, newly created positions, and positions of a limited duration, except the position of within the coverage of this Agreement, shall be posted on all applicable Bulletin Boards, for a minimum posting period of five (5) working days. During the job posting period, Permanent Full Time Employees will have the first opportunity to Page apply and be duly considered for such Job Posting. The provisions of Article shall not apply to vacancies. Should there be no successful applicants to the Job Posting from Permanent Full Time Employees, then second consideration shall be given to Regular Part Time Employees, third consideration shall be given to Probationary Full Time Employees, fourth consideration shall be given to Probationary Part Time, Part Time, and Temporary Employees, and fifth consideration shall be given to non-Employees. It is agreed and understood that Part Time, Probationaryand Temporary Employees have the opportunity to submit an application for a Job Posting when initially posted. It is agreed that an Employee cannot hold a Full Time and Part Time Job Posting, or two (2) Part Time Job under this Agreement, at the same time. Temporary vacancies, such as those caused by an Employee's absence due to an accident, injury, illness, sickness, vacation, leave of absence, suspension, or temporary transfer. shall not be posted. Such temporary vacancies may be filled at the discretion of the Employer for a period not to exceed twenty five (25) working days. The Employer, prior to assigning an Employee to such temporary vacancy shall give consideration to the most senior qualified Employee having regard for the immediate efficiency of Employer operations. If a temporary vacancy continues to exist beyond twenty five (25) working days, the vacant position will then be posted and filled in accordance with Article Contents The Job Posting notice shall contain the following information: job classification, initial section, initial reporting depot, minimum qualifications, shift and wage rate. A Job Posting notice as hereinbefore mentioned shall be in conformance with the Job Description and qualifications as previouslyagreed upon between the Employer and Union. Any changes in duties or qualifications will be discussed with the Union prior to implementation.

  • Local Circuit Switching Capability, including Tandem Switching Capability 4.2.1 Local circuit switching capability is defined as: (A) line-side facilities, which include, but are not limited to, the connection between a loop termination at a main distribution frame and a switch line card; (B) trunk-side facilities, which include, but are not limited to, the connection between trunk termination at a trunk-side cross-connect panel and a switch trunk card; (C) switching provided by remote switching modules; and (D) all features, functions, and capabilities of the switch, which include, but are not limited to: (1) the basic switching function of connecting lines to lines, line to trunks, trunks to lines, and trunks to trunks, as well as the same basic capabilities made available to BellSouth’s customers, such as a telephone number, white page listings, and dial tone; and (2) all other features that the switch is capable of providing, including but not limited to customer calling, customer local area signaling service features, and Centrex, as well as any technically feasible customized routing functions provided by the switch. Any features that are not currently available but are technically feasible through the switch can be requested through the BFR/NBR process.
