Histology. After experiments were completed, a 20–40 µA current was passed through each recording tetrode for 20-40 s while rats were under anesthesia immediately prior to euthanizing the rat, with the resulting brain lesions serving as confirmation of tetrode position. Transcardial perfusions were performed with 0.9% saline followed by 4% formalin. Brains were extracted and allowed to sit for several days in 4% formalin solution. Brains were moved to a 40% sucrose solution for approximately 72 hours, until brains sank to the bottom of the container, at which point brains were sliced into approximately 70 µm coronal slices and mounted on glass microscope slides. Brains were left for several days to dry in an 37° C oven, then Nissl stained with a cresyl violet solution.
Histology. Renal grafts were harvested in 10% formalin. Formalin fixed tissue was processed using a Leica ASP200 tissue processor; and embedded in wax. Sections (4µm) were cut using a microtome (Leica, Wetzlar, Germany), and baked overnight at 60ºC. Periodic acid Schiff’s staining (PAS) was performed by removing wax in xylene and rehydration through dips in 100% followed by 90% ethanol, and washing in running tap water for 5 minutes. Sections were incubated in 1% periodic acid for 10 minutes followed by washing with distilled water. Sections were then incubated in Schiff’s reagent (VWR) for 10 minutes followed by washing in running tap water for 5 minutes. Sections were then incubated in Xxxxx’x haematoxylin (VWR) for 3 minutes, followed by washing in running tap water for 10 minutes. Sections were then dehydrated through dips in 90% followed by 100% ethanol and xylene. Slides were mounted in DPX (VWR).

Histology. Atrial appendages (AA) were fixed in formalin and subse- quently embedded in paraffin. Two-micrometer sections were sliced perpendicular to the atrial wall, thus leaving a donut- shaped slice with full endocardial and epicardial circumfer- ence, and stained with Masson trichrome for microscopic examination. Section processing and analysis was performed by an investigator blinded to clinical data. Sections were analyzed using a 3 CCD RGB camera (Sony 950P, Sony, Japan) on a light microscope (Axiophot, Carl Zeiss, Germa- ny). Using a ×20 magnification objective (specimen level pixel size 0.64×0.64 μm2), tissue samples were systematical- ly measured going all around the section. Twelve to 25 mag- nification fields were analyzed per section. For each magnifi- cation, white areas (no tissue, lumen), the fibrotic epicardial and endocardial layers as well as areas with perivascular fibrosis, were semi-automatically excluded. Quantitative anal- ysis of interstitial fibrosis was performed by automatic color segmentation image analysis which automatically extracts green/blue color areas representing interstitial fibrosis. See Fig. 1. for a typical example. The volume percentage of collagen was expressed as the total area of fibrosis divided by the total test field minus white areas, epicardium, endocar- dium, and perivascular fibrosis. The number of fibrotic strings per magnification field (×20) was scored by two independent observers and divided into three categories: diffuse fibrosis=- score 0, one to two fibrotic strings=score 1, and more than two fibrotic strings=score 2.
