Specifically, the relative abundance of different vegetative compatibility types of the pathogenic fungus Cryphonectria parasitica and the frequency of hypovirulent strains will be assessed. These data will be correlated with the composition of the microbiome obtained by 16S gene sequencing. Finally, the effects of co-culture with bacteria isolated from the bark on the growth of the hypovirulent fungus will be evaluated. The methodologies that will be used are as follows: Field sampling; Isolation of fungus; Extraction of genomic DNA for genotypic characterisation of the fungus; Extraction of DNA from environmental samples; Bioinformatic analysis of metagenomics data; Isolation of bacteria with selective media and co-culture experiments with fungi. The results, in addition to having scientific relevance in the field of phytopathology, will be useful for implementing biological control strategies for chestnut blight. The data must be organized according to a precise and homogeneous archiving protocol which will subsequently allow the creation of a database. At the same time, the research fellow will have to reconstruct the lateral carbon flows of the agroecosystem in order to arrive at the determination of the net ecosystem balance of CO2 of the system itself, in the years in which the measurements are available.
