As a consequence, mBRCAtumors are vulnerable to drugs that introduce replication- associated DNA damage (platinum derivatives) or inhibit DNA repair pathways other than HR (such as PARP inhibitors - PARPi). Tumors that share molecular features of mBRCA tumors may also respond to similar therapeutic approaches (BRCAness). However, several mechanisms of resistance impairing the effectiveness of these drugs have been identified. Here, we will take clinical (profiling of pancreatic, breast, and ovarian cancers) and preclinical (using innovative patient- derived models) approaches to identify novel predictive biomarkers, develop synergistic therapeutic combinations and overcome resistance. The expected results of the proposed project will enhance our understanding of mBRCA biology and pave the way for the next generation of mBRCA-targeted therapeutics.
